Which material sounds better for speakers construction? Wood, Ply or MDF?
The title of your post should be "Which material sounds less". To that end and given the 3 choices it is MDF hands down.

Speaker cables if your amp isn't between your speakers
Get some Canare 4S11 and roll your own. Make them the length you need for each side, it will not effect the sound. Good luck. 
is it ok to leave a tube amp on without speakers connected
With tube amps the answer is NO!.you  will fry the output xfmr. SS amps may  be left on though however connecting and unconneti g spkr cables leaves you vulnerable to accidentally shorting the leads.

Yansad, if you haven't already done so and your domicile is conducive then spend money treating your room. 
Inexpensive speaker cables
Canare 4S11. Outstanding bar none. You can buy it by the foot at Markertek and roll you're own or there are EBay sellers who offer ready made sets. Great for biwiring also.
